Mechanism of Action
Acyclovir is an antiviral agent that works by:
 Killing or inhibiting the growth of viruses by interfering with their DNA replication.

Dosage
 Active Ingredient: Acyclovir
 Dose and duration should be determined by a physician, especially in children, elderly,
or those with renal impairment.
Side Effects
May include:
 Allergic reactions: Anaphylaxis, rash, urticaria
 Neurologic: Headache, dizziness, confusion, agitation, hallucinations, psychosis,
somnolence, seizures (especially in the elderly or those with renal impairment)
 Gastrointestinal: Nausea, diarrhea, gastrointestinal discomfort
 Liver: Elevated liver function tests
 Hematologic: Hemolysis, leukopenia, lymphadenopathy, disseminated intravascular
coagulation
 Skin: Alopecia, erythema multiforme, Stevens-Johnson syndrome, toxic epidermal
necrolysis
 Other: Fever, myalgia, peripheral edema
Drug Interactions
Use caution when co-administering with:
 Probenecid
 Cimetidine
 Mycophenolate mofetil
 Cyclosporin
 Tacrolimus
 Theophylline
Indications
 Treatment and prevention of:
o Chickenpox
o Shingles
o Cold sores
o Genital herpes
o Herpes simplex infections (initial and recurrent)
o Suppression of herpes virus in immunocompromised patients
Contraindications (When Not to Use)
 Hypersensitivity to acyclovir, valacyclovir, or any excipients in the formulation
Precautions
 Elderly patients and those with renal impairment require dose adjustment due to
reduced renal clearance.
 Monitor for neurological side effects in these populations; symptoms are generally
reversible upon discontinuation.
Warnings
Warning 1
Acyclovir is eliminated via the kidneys. Dose adjustment is essential in patients with renal
impairment.

May cause drowsiness or dizziness, which can impair the ability to drive or operate
