How It Works
Esomeprazole is a proton pump inhibitor (PPI) that binds irreversibly to the H⁺/K⁺ ATPase
enzyme in gastric parietal cells. This enzyme is responsible for the final step in acid secretion.
By blocking it, esomeprazole significantly reduces gastric acid production, increasing gastric pH
and promoting healing.

When Not to Use

Precaution
 Long-term use (e.g. >3 years) may impair vitamin B12 (cyanocobalamin) absorption,
leading to deficiency.
Warnings
Warning 1:
Hypomagnesemia (low magnesium levels) can occur with prolonged treatment. Monitor serum
magnesium levels periodically.
Warning 2:
Chronic PPI use may increase risk of osteoporosis-related fractures (hip, wrist, spine). Patients
at risk should follow bone health guidelines.
Warning 3:
Avoid concurrent use with clopidogrel unless necessary, due to potential reduction in
clopidogrel effectiveness.
