How It Works:
Pregabalin reduces neuronal calcium currents by binding to the alpha-2-delta subunit of
voltage-gated calcium channels in the central nervous system (CNS). This action is thought to
modulate the release of neurotransmitters and is responsible for its effects in neuropathic pain,
anxiety, and other pain syndromes.

Very Common:
Dizziness, drowsiness, somnolence
Common:
Increased appetite, euphoric mood, confusion, irritability, decreased libido,
disorientation, insomnia, ataxia, abnormal coordination, tremor, slurred speech
(dysarthria), memory impairment, difficulty concentrating, paraesthesia, sedation,
balance disorders, lethargy, headache, blurred vision, double vision (diplopia), vertigo,
vomiting, dry mouth, constipation, flatulence, erectile dysfunction, abnormal gait, feeling
drunk, fatigue, peripheral and general edema, weight gain
Uncommon:
Nasopharyngitis, anorexia, hypoglycemia, hallucination, panic attacks, restlessness,
agitation, depression, mood swings, depersonalization, difficulty finding words, abnormal
dreams, increased libido, anorgasmia, apathy, syncope, stupor, myoclonus, psychomotor
hyperactivity
Drug Interactions:
CNS depressants such as opiates, benzodiazepines
Ethanol (alcohol)
Lorazepam
Oxycodone
Indications:
Neuropathic pain associated with diabetic peripheral neuropathy
Management of postherpetic neuralgia
Pain associated with spinal cord injury
Adjunctive therapy in adults with partial seizures (with or without secondary
generalization)
Treatment of Generalized Anxiety Disorder (GAD) in adults
Treatment of fibromyalgia syndrome (FMS)
When Not to Use:
Known hypersensitivity to pregabalin or any component of the product
Patients with rare hereditary problems such as galactose intolerance, Lapp lactase
deficiency, or glucose-galactose malabsorption
Precautions:
Pregabalin should be withdrawn gradually over a minimum of 1 week to minimize the
risk of increased seizure frequency in patients with seizure disorders.
Warnings:
1. Discontinue immediately if signs of angioedema occur (swelling of face, lips, tongue,
throat).
2. Weight gain related to pregabalin dose and duration; diabetic patients may need to adjust
hypoglycemic medication accordingly.
3. May cause dizziness and somnolence — caution advised when driving or operating
machinery, especially in elderly patients.
